Microelution plates enable high-throughput TB-500 fragment detection in urine with comparable results to cartridges

Solid-phase extraction of small biologically active peptides on cartridges and microelution 96-well plates from human urine.

Background

Detecting the illicit use of small biologically active peptides like growth hormone releasing peptides (GHRPs), desmopressin, LHRH, and the short fragment of TB-500 is a critical task in anti-doping analysis. The current standard method involves liquid chromatography-mass spectrometry (LC-MS) following solid-phase extraction (SPE) on weak cation-exchange cartridges. While effective, this cartridge-based approach is often manual and time-consuming, limiting the number of samples that can be processed efficiently. This bottleneck poses a challenge for large-scale screening. To address this, researchers are exploring methods to increase throughput without sacrificing analytical performance, leading to the investigation of microelution plates which can handle many samples in parallel.

Study Design

This study evaluated a high-throughput sample preparation method for anti-doping analysis. They compared the standard weak cation-exchange cartridge SPE protocol with a modified protocol using weak cation exchange microelution 96-well plates. Human urine samples were diluted with 200 mM phosphate buffer to adjust pH. The extraction on microelution plates was performed using a Waters Positive Pressure-96 Processor. The effectiveness of the two protocols was assessed by comparing recovery, linearity, and reproducibility for 17 small bioactive peptides. The new protocol was also validated by analyzing urine samples known to contain ipamorelin, GHRP-6, and its metabolites.

Results

The microelution plate method demonstrated recovery, linearity, and reproducibility comparable to the established cartridge SPE technique. Diluting urine samples with 200 mM phosphate buffer provided sufficient buffering capacity without negatively impacting peptide recovery. The use of microelution plates successfully enabled the simultaneous processing of up to 96 samples, a significant increase in throughput. However, this high-throughput method came with a trade-off. The smaller sample volume used in the microelution plates reduced the pre-concentration factor.

This resulted in increased limits of detection for 5 out of the 17 peptides analyzed, making the method slightly less sensitive for those specific compounds.

Despite this, the protocol was successfully confirmed by analyzing real-world samples containing ipamorelin and GHRP-6. The study concludes that the new method is suitable for faster sample preparation and screening, while the traditional cartridge SPE with manual pH adjustment remains the superior choice for confirmatory analysis where maximum sensitivity is required.

Key Findings

  • Microelution plates allow for simultaneous processing of up to 96 urine samples.
  • The high-throughput method showed comparable recovery, linearity, and reproducibility to cartridge SPE.
  • The limits of detection increased for 5 out of 17 peptides due to a lower pre-concentration factor.
  • Dilution with 200 mM phosphate buffer effectively adjusted pH without harming peptide recovery.
  • Cartridge SPE with manual pH adjustment remains the best option for confirmation analysis.

Why It Matters

For anti-doping agencies, this research provides a validated method to significantly increase the speed and efficiency of screening for peptides like the TB-500 fragment. The ability to process up to 96 samples simultaneously reduces solvent consumption and labor, allowing for broader and more frequent testing. For athletes or individuals subject to testing, this means faster turnaround times for screening results. The key takeaway is that while this microelution method is excellent for initial high-volume screening, the classic, more sensitive cartridge method remains the gold standard for confirming a positive result. This establishes a two-tiered approach: a rapid, wide-net screening process followed by a more rigorous, sensitive confirmation for any flagged samples. This balanced strategy enhances the overall efficiency and reliability of anti-doping controls.
