Randomized trial to evaluate weekly dulaglutide, semaglutide, and tirzepatide in non-diabetic obese adults
Background
Obesity is a major public health challenge, often associated with metabolic dysfunction and increased risk of comorbidities. While GLP-1 receptor agonists (GLP-1RAs) have demonstrated significant efficacy in weight management and improving metabolic markers, initially in type 2 diabetes and now increasingly for obesity without diabetes, direct comparative data on the newer weekly GLP-1RAs is still evolving. Specifically, a head-to-head comparison of dulaglutide, semaglutide, and the dual GIP/GLP-1 receptor agonist tirzepatide in non-diabetic obese populations is needed to guide treatment selection.
Study Design
This randomized controlled trial plans to enroll 150 participants who are obese (with BMI >30) and do not have diabetes. Participants will be randomized into three distinct groups. Each group will receive one of three weekly GLP-1RAs: dulaglutide, semaglutide, or tirzepatide. The study will follow participants for a duration of 6 months. Primary endpoints include assessing changes in body weight, BMI, lipid profile, and liver functions to compare the efficacy of these agents.
Why It Matters
Directly comparing these potent GLP-1RAs in a non-diabetic obese cohort is crucial for optimizing treatment strategies and personalizing care. The findings from this trial will provide valuable evidence for clinicians and individuals, helping them make informed decisions about which weekly GLP-1RA might offer the best balance of efficacy for weight loss and metabolic improvements in the absence of diabetes. This could lead to more tailored prescribing practices, potentially influencing protocol selection for individuals seeking significant weight management. Understanding the comparative profiles of dulaglutide, semaglutide, and tirzepatide could refine current obesity treatment guidelines and inform future drug development.
