GHRP-2 Shows Robust Growth Hormone Release in Adolescents

Background

Growth hormone-releasing peptide 2 (GHRP-2) is a synthetic peptide that potently stimulates the release of growth hormone (GH) from the pituitary gland by activating the ghrelin receptor. GH is a critical hormone for normal growth, metabolism, and development, especially during the rapid growth phase of adolescence. While the GH-releasing effects of GHRP-2 are established in adults, its specific efficacy and safety profile in a healthy adolescent population, a key demographic for growth-related studies, have been less comprehensively characterized.

Results

Administration of GHRP-2 resulted in a rapid and statistically significant elevation of serum GH levels compared to the placebo group. The mean peak GH concentration observed in the GHRP-2 group was 28.5 ± 4.2 ng/mL, representing a remarkable 5.3-fold increase from baseline levels, whereas the placebo group exhibited no significant change (p<0.001). The area under the curve (AUC) for GH levels over the 180-minute observation period was 3200 ± 350 ng·min/mL in the GHRP-2 group, which was 43% higher than the placebo group's AUC (p<0.0001), clearly demonstrating a robust and sustained GH secretagogue effect. Furthermore, 88% of adolescents in the GHRP-2 group showed a peak GH response exceeding 10 ng/mL, a threshold often used in diagnostic testing. The peptide was generally well-tolerated, with only mild, transient side effects such as flushing reported in 15% of the GHRP-2 recipients, and no serious adverse events occurred.

Why It Matters

This study provides compelling evidence for the potent growth hormone-releasing activity of GHRP-2 in healthy adolescents, confirming its strong pharmacological effect in this age group. The robust and consistent GH response suggests GHRP-2 could be a valuable diagnostic tool for assessing GH deficiency or a potential therapeutic agent for various growth disorders. Further research, including larger cohorts and long-term safety and efficacy assessments, is warranted to explore its clinical utility and could lead to Phase II trials investigating its therapeutic potential in adolescents with conditions like idiopathic short stature.
