Background

Opioid dependence and the associated morphine withdrawal syndrome present immense challenges in public health, often leading to relapse due to severe and distressing aversive symptoms. Current therapeutic strategies for managing withdrawal have limitations, highlighting a critical need for novel, effective, and non-opioid treatment options. This study investigates the potential of Selank, a synthetic peptide analog of tuftsin, to alleviate the acute aversive signs of morphine withdrawal.

Results

Selank demonstrated significant efficacy, reducing the total index of morphine withdrawal syndrome by 39.6% compared to the active control group. It specifically and significantly (p<0.0001) attenuated several key aversive signs, including convulsive reactions, ptosis, and posture disorders. The most striking finding was its impact on sensory hypersensitivity: Selank 9-fold increased the tactile sensitivity threshold in morphine-dependent rats, indicating a substantial reduction in the painful hypersensitivity often associated with opioid withdrawal. While highly effective, Selank was slightly inferior to diazepam (a benzodiazepine often used for withdrawal management), which reduced the total withdrawal index by 49.3% and increased the tactile sensitivity threshold 13-fold.

This research suggests that Selank could be a promising novel therapeutic candidate for managing the distressing and often debilitating symptoms of opioid withdrawal, offering a potential non-opioid alternative.
