Selank alone shows no effect on GABAergic gene expression but modifies effects of GABA and olanzapine in IMR-32 cells
Background
The peptide Selank is known for its anxiolytic effects, which clinical studies suggest are comparable to classical benzodiazepine drugs. Benzodiazepines work by enhancing the inhibitory effect of the neurotransmitter GABA through allosteric modulation of GABAA receptors. This clinical observation led to the hypothesis that Selank's molecular mechanism might also involve modulating the GABAergic system. The GABAergic system is the primary inhibitory neurotransmission system in the brain, playing a crucial role in regulating neuronal excitability and anxiety. Understanding how Selank interacts with this system at the genetic level could clarify its mechanism of action and its potential as a therapeutic agent.
Study Design
This in vitro study investigated changes in the expression of 84 genes involved in GABAergic neurotransmission using human neuroblastoma IMR-32 cells. The researchers used the qPCR method to measure mRNA levels. The cells were treated with several test substances: GABA, the peptide Selank, the atypical antipsychotic olanzapine, and two combinations: Selank + GABA and Selank + olanzapine. The primary endpoint was the change in mRNA expression levels of the target genes following exposure to these compounds, compared to untreated control cells.
Results
The study found that Selank, when administered alone, produced no changes in the mRNA levels of the 84 GABAergic system genes studied in the IMR-32 cell line. In contrast, GABA alone did alter the expression levels of certain genes. The combination of Selank and GABA led to a nearly complete suppression of the gene expression changes that were induced by GABA alone. When Selank was co-administered with olanzapine, it amplified the drug's effects.
The number of genes with altered expression was greater with the Selank + olanzapine combination compared to the number affected by olanzapine alone. These results suggest that while Selank doesn't directly regulate the transcription of these specific genes, it does modulate the cellular response to other GABAergic agents. The findings partially support the hypothesis that Selank may influence the interaction of GABA with its
GABAAreceptors.
Key Findings
- Selank alone had no direct effect on the mRNA levels of 84 GABAergic system genes in IMR-32 cells.
- The combined application of Selank and GABA resulted in the nearly complete suppression of GABA-induced gene expression changes.
- When combined with olanzapine, Selank led to expression changes in more genes than olanzapine did alone.
- The data partially confirms the hypothesis that Selank may affect the interaction of GABA with GABAA receptors.
Why It Matters
This research suggests Selank's mechanism isn't a direct up- or down-regulation of GABAergic gene expression, but rather a modulatory one. For users, this implies Selank may be more effective as an adjunct or potentiator rather than a standalone agent for directly altering GABAergic tone at the genetic level. The peptide appears to normalize or buffer the system's response to GABA and enhance the effects of other drugs like olanzapine. This points to potential stacking protocols where Selank could be used to amplify the therapeutic effects of certain pharmaceuticals or to stabilize the GABA system's response to its primary ligand. Further research is needed to confirm these interaction effects in vivo and determine optimal combination strategies and clinical relevance for conditions like anxiety or for mitigating side effects of antipsychotics.
