Epitalon and Melatonin Extend Lifespan, Reduce Cancer in Aging Mice

Background

Aging is a complex process characterized by progressive decline in physiological function and an increased susceptibility to age-related diseases, including cancer. Senescence-accelerated mice (SAM) are a well-established animal model that spontaneously develops various age-related pathologies and a shortened lifespan, making them ideal for studying anti-aging interventions. Both Epitalon (a synthetic peptide) and Melatonin (a natural hormone) have demonstrated anti-aging, antioxidant, and immunomodulatory properties in various models. This study aimed to investigate the individual and combined effects of Epitalon and Melatonin on lifespan extension and the prevention of spontaneous tumor development in SAM mice.

Results

The study revealed significant anti-aging and anti-carcinogenic effects for both compounds, particularly in combination. Epitalon treatment alone increased the median lifespan by 15% (from 12.5 months in controls to 14.4 months) and reduced overall spontaneous tumor incidence by 25% (p<0.05). Melatonin treatment similarly extended median lifespan by 10% (to 13.8 months) and decreased tumor incidence by 18% (p<0.05). The most pronounced effects were observed in the combination group: > The co-administration of Epitalon and Melatonin resulted in a remarkable 22% increase in median lifespan (extending it to 15.25 months) and a 35% reduction in spontaneous tumor incidence (p<0.001) compared to control mice. This synergistic effect was also evident in specific tumor types, with a 40% reduction in lymphoma incidence and a 30% decrease in lung adenomas in the combined treatment group.

Why It Matters

This research powerfully demonstrates the potential of Epitalon and Melatonin as effective geroprotectors (substances that protect against aging) and chemopreventive agents, especially when used in combination.
