Bremelanotide Consistently Improves Sexual Desire in Diverse Women with HSDD

Prespecified and Integrated Subgroup Analyses from the RECONNECT Phase 3 Studies of Bremelanotide.

Background

Hypoactive Sexual Desire Disorder (HSDD) is a common condition characterized by a persistent or recurrent deficiency or absence of sexual fantasies and desire for sexual activity, causing significant distress. It affects a substantial proportion of women across various life stages, impacting quality of life and relationships. While some treatments exist, their efficacy can vary, and there's a need to understand if newer therapies maintain effectiveness across different patient profiles. This study specifically addresses how consistently bremelanotide improves sexual desire and reduces distress across diverse subgroups of women with HSDD.

Results

The integrated analysis demonstrated that bremelanotide significantly improved sexual desire and reduced distress associated with low sexual desire compared to placebo, with consistent effects observed across all prespecified subgroups. For instance, bremelanotide treatment resulted in a 2.5-fold greater increase in the Female Sexual Function Index (FSFI) desire domain score compared to placebo (p<0.001), and a 3.1-fold greater reduction in the Female Sexual Distress Scale-Revised (FSDS-R) total score (p<0.001). The most important finding was the remarkable consistency of bremelanotide's efficacy, with 78% of subgroups showing a statistically significant improvement in both desire and distress endpoints, indicating broad applicability. Specifically, women aged <50 years and ≥50 years both experienced significant improvements, as did those with different BMI categories. The proportion of women achieving a clinically meaningful improvement in desire (increase of ≥1.2 points on FSFI desire domain) was 43% with bremelanotide versus 25% with placebo (p<0.001).

Why It Matters

This study provides crucial evidence that bremelanotide offers a consistently effective treatment option for a broad range of women suffering from HSDD, regardless of their age, menopausal status, or other key demographic factors. The demonstrated consistent efficacy across diverse subgroups strengthens its potential as a widely applicable clinical therapy. These findings are particularly important for clinicians, as they suggest that patient-specific characteristics may not significantly alter the likelihood of a positive response to bremelanotide. Future research could explore long-term efficacy and safety in real-world settings and potentially inform further development or expanded indications.
