Background

Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy characterized by an immunosuppressive and metabolically rewired tumor microenvironment (TME), contributing to therapeutic resistance. Current treatments often fall short due to this hostile environment. Alpha-enolase (ENO1) is frequently overexpressed in PDAC and linked to poor prognosis, yet its precise role in TME remodeling and its interaction with key immunosuppressive pathways like TGFβ activation and metabolic adaptation remains largely undefined. This study aimed to clarify ENO1's function and explore the therapeutic potential of targeting it.
