PET Radiotracers Targeting MDM2 Protein Interaction: From Preclinical Validation to First-in-Human Studies.
Background
Current imaging techniques struggle to accurately define surgical margins for well-differentiated liposarcoma (WDLS), leading to incomplete resections or overtreatment. The MDM2 oncoprotein, which inhibits the p53 tumor suppressor and is amplified in WDLS, presents a promising target for both diagnosis and therapy. Developing specific imaging probes for MDM2 could significantly improve surgical planning and patient outcomes by precisely delineating tumor boundaries and assessing MDM2 expression.
