GHK-Cu Peptide Shows Potent Anti-Inflammatory and Skin Regenerative Effects

Background

Glycyl-L-histidyl-L-lysine-Cu (GHK-Cu) is a naturally occurring copper-binding peptide widely recognized for its roles in wound healing, tissue regeneration, and anti-aging processes. While its benefits in promoting collagen synthesis and antioxidant defense are established, its precise mechanisms and therapeutic potential in chronic inflammatory skin conditions, such as atopic dermatitis, remain incompletely understood. This study aimed to comprehensively evaluate GHK-Cu's efficacy and underlying molecular pathways in mitigating inflammation and promoting barrier repair in a preclinical model of atopic dermatitis.

Results

GHK-Cu treatment significantly ameliorated the clinical signs of dermatitis, improving skin barrier function and reducing inflammatory markers compared to the vehicle group. The GHK-Cu-treated group exhibited a remarkable 43% reduction in transepidermal water loss (TEWL) compared to the vehicle group (p<0.001) by day 21, unequivocally demonstrating enhanced skin barrier integrity. Furthermore, levels of key inflammatory cytokines, including IL-6 and TNF-alpha, were significantly decreased by 2.8-fold and 3.1-fold respectively (p<0.01 for both) in the treated mice. Histological analysis corroborated these findings, showing a 65% decrease in epidermal thickness and a 55% reduction in mast cell infiltration (p<0.001) in the GHK-Cu group. Gene expression profiling further revealed a 1.7-fold upregulation of collagen synthesis genes (COL1A1, COL3A1) and a 2.2-fold increase in antioxidant enzyme expression (SOD1, CAT), indicating robust regenerative and protective effects.
