GRP78 protein binds to the MC4R, regulating receptor function and energy homeostasis in mice

Glucose-regulated protein 78 binds to and regulates the melanocortin-4 receptor.

Background

The melanocortin-4 receptor (MC4R) is a key G protein-coupled receptor (GPCR) that plays an essential role in regulating energy balance and body weight. Disruptions in MC4R signaling are linked to severe obesity. Understanding the proteins that interact with and regulate MC4R is critical for developing more effective treatments for metabolic disorders. This research explores the relationship between MC4R and other cellular proteins to uncover novel mechanisms that control its trafficking and signaling activity, potentially revealing new therapeutic targets for managing energy homeostasis.

Study Design

Researchers identified MC4R-interacting proteins using a pulldown assay with hypothalamic protein extracts and the third intracellular loop of MC4R. They investigated the interaction and its functional consequences in vitro by downregulating GRP78 with a short interfering RNA (siRNA) approach and inducing endoplasmic reticulum stress with tunicamycin. To assess in vivo relevance, they used a lentiviral-mediated short hairpin RNA (shRNA) to knock down endogenous GRP78 specifically in the paraventricular nucleus (PVN) of the hypothalamus in mice fed a high-fat diet.

Results

Glucose-regulated protein 78 (GRP78) was identified as a novel MC4R-interacting protein. This interaction occurs both in the cytosol and at the cell surface, and it intensifies when MC4R is internalized following stimulation with the agonist Melanotan-II (MTII). Downregulating GRP78 expression using siRNA significantly attenuated this MTII-mediated receptor internalization. Similarly, reducing GRP78 during tunicamycin-induced endoplasmic reticulum stress suppressed both MTII-mediated internalization of MC4R and its cAMP-mediated transcriptional activity. The study suggests GRP78 has a chaperone-like role in MC4R trafficking and signaling.

Furthermore, in vivo knockdown of endogenous GRP78 in the hypothalamic paraventricular nucleus (PVN) resulted in an increase in body weight in mice that were fed a high-fat diet.

Key Findings

  • Glucose-regulated protein 78 (GRP78) was identified as a novel MC4R-interacting protein.
  • Interaction between GRP78 and MC4R increases when MC4R is internalized in the presence of the agonist Melanotan-II.
  • Downregulation of GRP78 attenuated Melanotan-II-mediated MC4R internalization and cAMP transcriptional activity.
  • Knockdown of GRP78 in the hypothalamic PVN led to increased body weight in mice on a high-fat diet.

Why It Matters

This study reveals a previously unknown regulatory mechanism for MC4R, a critical receptor in weight management. The finding that GRP78 is essential for proper MC4R function suggests that cellular stress (which can affect GRP78 levels) might impair melanocortin signaling, potentially contributing to metabolic dysfunction. For biohackers or clinicians using MC4R agonists like Melanotan-II, this implies that overall cellular health and particularly endoplasmic reticulum function could influence the efficacy and response to these peptides. Optimizing cellular chaperone function could be a novel strategy to enhance MC4R-targeted therapies, although this concept is far from clinical application and requires extensive further research to translate into a usable protocol.
