PT-141: A Melanocortin Agonist for Hypoactive Sexual Desire
Background
PT-141 (Bremelanotide), an oligopeptide with a molecular formula of C50H68N14O10 and a molecular weight of 1025.2 g/mol, represents a novel therapeutic approach. This compound is designed to address hypoactive sexual desire disorder (HSDD), a common condition characterized by a persistent or recurrent deficiency or absence of sexual fantasies and desire for sexual activity, causing significant distress. Despite its prevalence, there has been a notable lack of effective, on-demand pharmacological treatments specifically targeting the neurobiological pathways underlying HSDD.
Results
The clinical trials demonstrated significant improvements in key measures of sexual function and distress compared to placebo. In the pivotal trials, 25% of women treated with PT-141 reported a clinically meaningful increase in their desire score (measured by the Female Sexual Function Index-Desire domain, FSFI-D), compared to 17% in the placebo group (p=0.001). Furthermore, 35% of PT-141-treated women experienced a clinically meaningful decrease in distress associated with low sexual desire (measured by the Female Sexual Distress Scale-Desire/Arousal/Orgasm, FSDS-DAO), versus 25% in the placebo group (p=0.0008). > The most important finding was that PT-141 significantly increased sexual desire and reduced the personal distress associated with low sexual desire in premenopausal women with HSDD, providing an on-demand treatment option. This represents a 1.5-fold greater likelihood of desire improvement and a 1.4-fold greater likelihood of distress reduction compared to placebo.
