Background

Growth hormone secretagogues (GHSs) like NN703 and ipamorelin are synthetic compounds that stimulate the body's natural release of growth hormone (GH). While promising for various therapeutic applications, optimizing their potency and bioavailability remains a challenge. This study addresses the knowledge gap in designing GHSs that combine high potency with desirable pharmacological properties by exploring novel hybrid structures.

Results

The re-introduction of certain pharmacophores (molecular features responsible for drug action) into the C-terminal region of NN703, which had previously been removed to improve oral bioavailability, led to a remarkable discovery. These hybrid compounds demonstrated unexpectedly high potency both in vitro and in vivo. They significantly enhanced growth hormone release, far exceeding the activity of their parent compounds. The in vivo pig study confirmed that a single dose of 50 nmol/kg was sufficient to elicit a potent growth hormone secretagogue effect. The strategic re-introduction of specific C-terminal pharmacophores resulted in hybrid compounds that were significantly more potent than previous NN703 optimizations, both in cellular assays and live animal models.
