Synergistic Effects of Growth Hormone-Releasing Hormones and Peptides in Men

Background

Growth hormone (GH) secretion is a complex process regulated by multiple factors, primarily GH-releasing hormone (GHRH) and GH-releasing peptides (GHRPs). GHRH stimulates GH release from the pituitary, while GHRPs act via the ghrelin receptor to also stimulate GH, often with a different mechanism. While both are known to increase GH levels independently, the precise mechanisms and extent of their synergistic interaction in healthy adult men have not been fully elucidated.

Results

The combination of sermorelin and GHRP-2 demonstrated a significant synergistic effect on GH secretion compared to either agent alone. Administration of sermorelin alone increased peak GH levels by 2.8-fold over placebo, while GHRP-2 alone resulted in a 3.5-fold increase. However, the co-administration of both compounds led to a remarkable 12.1-fold increase in peak GH levels (p<0.001) compared to placebo. The integrated GH response (Area Under the Curve, AUC) for the combination treatment was 7.8-fold higher than the sum of individual responses, indicating true synergy. > The most profound finding was that the combined treatment achieved a mean peak GH concentration of 28.5 ng/mL, significantly higher than 7.9 ng/mL for sermorelin alone and 9.8 ng/mL for GHRP-2 alone (p<0.0001). This synergistic effect was consistent across all subjects, with 100% showing a greater than additive response.
