Background

Metabolic dysfunction-associated steatotic liver disease (MASLD), formerly NAFLD, is a leading cause of chronic liver disease, affecting approximately 25% of adults globally. It progresses from simple steatosis to MASH, increasing risks of cirrhosis and cardiovascular mortality. Current treatments like resmetirom and semaglutide exist, but there's a critical need for complementary therapies with diverse mechanisms. Lipotoxicity, oxidative stress, and endoplasmic reticulum (ER) stress are key drivers of MASLD progression, particularly through activation of innate immune pathways.
