Background
Growth hormone (GH) and insulin-like growth factor I (IGF-1) are critical for metabolic regulation, body composition, and overall well-being. While GH-releasing hormone (GHRH) naturally stimulates GH secretion, its very short half-life limits its therapeutic utility for conditions like GH deficiency. This study aimed to determine if CJC-1295, a novel long-acting GHRH analog, could provide sustained stimulation of GH and IGF-1 secretion in healthy human adults.
Results
A single dose of CJC-1295 resulted in a rapid and significant increase in GH secretion, with peak levels observed within 2 to 4 hours post-administration. > The most important finding was that CJC-1295 provided a remarkably prolonged effect, sustaining elevated GH levels for up to 14 days and IGF-1 levels for over 10 days from a single injection. Specifically, the 60 mcg/kg dose led to a 2.5-fold increase in mean GH area under the curve (AUC) compared to placebo over 7 days (p<0.001), and IGF-1 levels increased by an average of 1.8-fold above baseline, maintained for 10-14 days. This sustained elevation was significantly superior to the transient effects of native GHRH, demonstrating a clear advantage in duration of action.
