Tirzepatide vs. Semaglutide Phase 3b Trial Initiated for Obesity and Overweight Without Type 2 Diabetes

Obesity and overweight with comorbidities represent a significant global health challenge, increasing risks for cardiovascular disease, certain cancers, and metabolic dysfunction. Current weight management strategies, including lifestyle interventions and existing pharmacotherapies, often achieve modest or unsustainable weight loss for many individuals. Glucagon-like peptide-1 receptor (GLP-1R) agonists have revolutionized treatment, demonstrating substantial efficacy. The emergence of dual GLP-1R/GIPR agonists like tirzepatide offers potentially superior weight loss by targeting multiple incretin pathways, warranting direct comparison against established GLP-1R monotherapy.

This Phase 3b randomized, double-blind study aims to compare the efficacy and safety of tirzepatide against semaglutide in adult participants with obesity or overweight who have weight-related comorbidities but without Type 2 Diabetes. Participants will undergo treatment for approximately 74 weeks. The primary objective is to evaluate the comparative effectiveness of these two leading incretin-based therapies in a head-to-head setting, focusing on weight reduction and safety profiles in a non-diabetic population.
