Mazdutide Plus Lifestyle Intervention Trial for Obesity Weight Management

ASCEND-1: Lifestyle Intervention Plus Mazdutide for Weight Management

Background

Obesity is a complex, chronic disease affecting millions globally, significantly increasing the risk of conditions like type 2 diabetes, cardiovascular disease, and certain cancers. While lifestyle interventions are foundational, achieving and maintaining sustained weight loss often proves challenging for many individuals. This upcoming clinical trial aims to address this critical knowledge gap by evaluating whether combining a novel drug, Mazdutide, with an intensive lifestyle intervention can lead to more effective and durable weight management outcomes.

Study Design

Population
Participants with obesity.
Intervention
Mazdutide combined with intensive lifestyle intervention for 52 weeks.
Comparator
Lifestyle intervention alone.
Outcome
Mean percentage change in body weight from baseline.

Results

This RANDOMIZED, QUADRUPLE, TREATMENT trial is currently RECRUITING participants, with an estimated completion in December 2028, meaning no results are available yet. The study is meticulously designed to investigate whether the combination of Mazdutide, a novel dual GLP-1/glucagon receptor agonist (a type of drug that mimics natural hormones to regulate metabolism and appetite), and intensive lifestyle intervention can achieve superior and sustained weight control compared to lifestyle intervention alone. Researchers hypothesize that participants receiving Mazdutide will demonstrate a significantly greater mean percentage of body weight loss from baseline over the 52-week intervention period. > The primary outcome will assess the mean percentage change in body weight from baseline, with an expectation of a clinically meaningful reduction (e.g., >10% of initial body weight) in the active treatment group. Secondary outcomes will include evaluating improvements in key metabolic parameters such as fasting blood glucose, HbA1c, lipid profiles (cholesterol, triglycerides), and blood pressure, as well as assessing changes in body composition and quality of life metrics.
