New Dried Blood Spot Method Boosts GHRP-2 Doping Detection Window

On the road of dried blood spot sampling for antidoping tests: Detection of GHRP-2 abuse.

Background

Growth Hormone-Releasing Peptides (GHRPs), such as GHRP-2, are potent secretagogues that stimulate the body's natural production of growth hormone (GH). These substances are frequently abused in sports for their anabolic and recovery-enhancing effects, leading to unfair advantages and health risks. Traditional anti-doping tests often struggle with a short detection window for these rapidly metabolized compounds, creating a significant challenge for regulatory bodies. This study addresses the urgent need for improved analytical methods to extend the detection period of GHRP-2 abuse.

Results

The developed LC-MS/MS method demonstrated exceptional analytical performance, achieving a limit of detection (LOD) of 0.05 ng/mL and a limit of quantification (LOQ) of 0.1 ng/mL for GHRP-2 in DBS, indicating high sensitivity. Recovery rates were consistently robust, ranging from 90% to 105% across all tested concentrations, confirming efficient extraction from the DBS matrix. > The study successfully extended the detection window for GHRP-2 to up to 72 hours post-administration in DBS samples, representing a 2-fold increase compared to typical plasma detection times of 24-36 hours. Specificity was confirmed with no significant interference from endogenous compounds or other common doping agents, showing a 99.5% specificity rate, ensuring reliable identification of GHRP-2 abuse.

Why It Matters

This study's findings represent a significant advancement in anti-doping science, offering a more effective tool for detecting GHRP-2 abuse. The extended detection window provided by DBS sampling makes it considerably harder for athletes to evade testing, thereby promoting fairer competition. Furthermore, DBS collection is minimally invasive, easier to transport, and more stable than traditional blood samples, simplifying logistics for anti-doping agencies. This method has strong potential for rapid implementation into routine anti-doping programs, potentially leading to more accurate and widespread testing. Future steps include large-scale validation in diverse populations and integration into international anti-doping protocols.
