Background
Monogenic obesity is a rare, severe form of obesity characterized by early onset and significant weight gain, driven by genetic alterations in critical hypothalamic pathways, particularly the leptin/melanocortin pathway. Over 60 genes encoding proteins within this pathway have been identified. Patients with monogenic obesity face severe complications and a major impact on their quality of life. Current management strategies, including traditional lifestyle interventions and bariatric surgery, often show disappointing results with frequent weight regain, highlighting a critical need for more effective therapeutic approaches.
