Semax and Melanotan II Show Antidepressant-Like Effects in Stressed Rats

Background

Chronic stress and major depressive disorder (MDD) are widespread conditions, often resistant to current treatments, highlighting an urgent need for novel therapeutic strategies. The hypothalamic-pituitary-adrenal (HPA) axis plays a crucial role in stress response, and its dysregulation is implicated in depression. Results

Both Semax and Melanotan II demonstrated significant antidepressant-like and antistress effects. In the forced swim test, Semax treatment led to a remarkable 42% reduction in immobility time compared to the stressed control group (p<0.01), indicating improved coping mechanisms. Melanotan II similarly reduced immobility by 38% (p<0.05). Both compounds also significantly increased exploratory behavior in the open field test, with Semax showing a 2.3-fold increase and Melanotan II a 1.9-fold increase in distance traveled compared to stressed controls (p<0.05 for both). The most striking finding was that both Semax and Melanotan II normalized stress-induced elevations in plasma corticosterone levels by approximately 30-35% (p<0.01), bringing them closer to levels observed in non-stressed animals.

These findings are significant as they identify Semax and Melanotan II as promising candidates for treating stress-related disorders and depression, potentially offering a new class of neuroactive peptides.
