Novel Peptide Prevents Fatty Liver Disease in Mice Fed High-Fat Diet

Background

Non-alcoholic fatty liver disease (NAFLD) is a growing global health concern, characterized by excessive fat accumulation in the liver, often progressing to non-alcoholic steatohepatitis (NASH) and cirrhosis. It is strongly associated with obesity, type 2 diabetes, and metabolic syndrome. Current therapeutic options for NAFLD are limited, highlighting an urgent need for effective preventive strategies, particularly those leveraging nutritional interventions. This study specifically aimed to investigate the preventive potential of a novel food-derived peptide, HepatoGuard-1, against high-fat diet-induced NAFLD in a preclinical model.

Results

Mice receiving HepatoGuard-1 showed significantly attenuated NAFLD development compared to the unsupplemented HFD group. The 10 mg/kg HepatoGuard-1 group exhibited a 45% reduction in liver triglyceride content (p<0.001) and a 38% decrease in liver weight (p<0.01) compared to HFD controls. Histopathological analysis revealed a 60% reduction in hepatic steatosis scores and a 55% decrease in inflammatory cell infiltration in the high-dose treatment group (p<0.001). Gene expression analysis showed a 2.3-fold upregulation of antioxidant enzymes (Nrf2) and a 1.8-fold downregulation of pro-inflammatory cytokines (TNF-α, IL-6) in the livers of treated mice (p<0.05).

Why It Matters

This study provides compelling evidence that HepatoGuard-1 has significant preventive effects against high-fat diet-induced NAFLD and associated metabolic dysfunction. The findings suggest that this novel food-derived peptide could represent a promising nutritional strategy for the prevention and early intervention of NAFLD. If these results translate to human studies, it could lead to the development of functional foods or dietary supplements for individuals at high risk of NAFLD. Future research should focus on elucidating the precise molecular mechanisms and conducting Phase I and Phase II human clinical trials to confirm safety and efficacy.
