New Method Improves Detection of Prohibited Growth Hormone Releasing Hormone Analogs

Background

The administration of growth hormone releasing hormone (GHRH) and its synthetic analogs is strictly prohibited by the World Anti-Doping Agency (WADA) due to their performance-enhancing potential. Despite intelligence and admissions of use, these substances are rarely detected in anti-doping samples by WADA-accredited laboratories. This challenge arises from their typically small concentration in urine and limited knowledge about their metabolism, especially for unapproved synthetic analogs, making them difficult to identify.

Results

The research successfully identified and characterized 19 major in vitro metabolites across the four GHRH synthetic analogs, providing critical new targets for anti-doping analysis. The developed LC-MS/MS method demonstrated high sensitivity for these compounds. The limits of detection (LODs) for the target peptides were generally 1 ng/ml or less, which successfully meets or exceeds the WADA required performance limit for anti-doping samples. This enhanced sensitivity means that even low concentrations of these prohibited substances can now be reliably detected. The comprehensive characterization of these metabolites offers an invaluable resource for anti-doping laboratories worldwide, enabling more accurate and robust testing.
